Resorufin acetate is a fluorometric probe that acts as a substrate for cytosolic aldehyde dehydrogenase (ALDH1A1) esterase activity. It is also a useful substrate for chymotrypsin. Upon enzymatic cleavage, the resorufin anion displays red fluorescence (ex/em maxima = 570/580 nm, respectively).

Resorufin acetate is a cytosolic fluorescent dye used in the detection of bacterial enzyme activity. It is also used to measure the metabolic rate of bacteria, yeast, and mammalian cells. Resorufin acetate binds to acyl chains and causes a shift in the ph profile of the cell. This dye also reacts with disulfide bonds in proteins to form resorufin-disulfide products. The nmr spectra of resorufin acetate show that it has a phenoxazine group and a carbonyl group.
