Fluorescein diacetate

3,6-Diacetoxyfluoran; Di-O-acetylfluorescein

Widely-used fluorogenic substrate for esterase activity. Upon cleavage fluorescein is released producing a green fluorescence that can be read using standard FITC filter sets. Common viability stain for plant cells, protoplasts and tissues.

Diacetylfluorescein is used in fluorometric staining in the imaging of mammalian cells. Also used in the synthesis of nanocrystal biolabels with releasable fluorophores used in immunoassays.

Fluorescein diacetate is a fluorescent dye with biochemically active properties. It can be used as a marker for mitochondrial functions and nuclear DNA, and has been shown to inhibit the polymerase chain reaction. Fluorescein diacetate is also effective in inhibiting the growth factor-β1 activity of cells in culture, which may be due to its ability to bind to cellular nuclei. This compound has also been shown to have an antitumor response against HL-60 cells, which may be due to its ability to penetrate the cell membrane and inhibit cellular protein synthesis.
